Is there any way to have a constraint be tied to a specific day of the week?
For example, I have a task that needs to be performed on a weekend in order to be the least disruptive.
Or would a better option be to create a custom calendar for the resource assigned to the task that would only show weekend availability?

I would go with the calendar for the resource assigned to the task. You can create a task calendar and assign it to the task if that is better for you, I feel that just adds another level of calendaring to keep track of.
